Quantitative Determination of 4 Fat-soluble Vitamins and Cholesterol in Food by Liquid Chromatography and Gas Chromatography-Tandem Mass Spectrometry

A LC/MS/MS method for the simultaneous determination of 4 fat-soluble vitamins including vitamin A (retinal), vitamin D2 (ergocalciferol), vitamin D3 (Cholecalciferol) and vitamin E (Tocopherol-_) in general food was developed and validated. GC/MS/MS was applied to analyze cholesterol. Sample was weigh (5 g) and saponified by 7.5% KOH methanol solution for 60 min. The saponified test portion was extracted by n-heptane:n-hexane (1:1, v/v). After dryness with rotary evaporators, the substrate was reconstituted with Tetrahydrofuran.

Recoveries were located in the range of 91–109%. The developed approach has been applied successfully for efficient determination of 4 fat-soluble vitamin and cholesterol in food materials.
