Quantitative Determination of Seven Tetracyclines in Fish Muscles by Liquid Chromatography-Tandem Mass Spectrometry

A confirmatory method based on liquid chromatography-tandem mass spectrometry (LC/MS/MS) has been developed for the low-level determination of residues of 7 kinds of tetracyclines in fish muscle e.g., tetracyclines, oxytetracycline, chlortetracycline, doxycycline, 4-epimer-tetracycline, 4-epimer-oxytetracycline and 4-epimer-chlortetracycline. The procedure contains solvent extraction by McIlvaine (10 mM EDTA, pH=4.0), followed by solid phase extraction with HLB column, LC/MS/MS analysis by positive electrospray ionization (ESI) was applied multiple reaction monitoring of two transition reactions for each compound. Average fortification recoveries of the 7 compounds tested in fish muscles were 94%, 104%, 100%, 102%, 105%, 98% and 103%, respectively. In fish muscle, the decision limits (CCα) were 0.35-0.99 μg/kg, and the detection capabilities (CCβ) 0.38-1.16 μg/kg (n=15), thus below the minimum required performance limit (MRPL) set at 2.5  μg/kg in fish muscles by Taiwan FDA.
